Will the real guest that booked please stand up

On my second booking for my house, I have not heard back from the booking guest except at initial booking. His sister contacted me to my cell from her cell wanting to know when he will get keys but doesn't know when he will arrive. What do I do if he doesn't show up at check in. Can I refuse, do I have to wait for him? Help please.
