We are setting up a four bedroom two bath holiday rental on The Greek island of Rhodes. There are two bedrooms and a bathroom upstairs and the same downstairs. My question is do you recommend we have one adult room with one large bed and the other with two twin beds on each floor to accommodate families? I imagine kids don’t want to share a bed and often times two families will travel together in Europe, so might it be good to have a kids room for each family?

Invest in single beds that can be put together so 2 couples can stay. Best of both worlds.

@Villa-Barbara0  This is a great idea and very thoughtful and accommodating for families. If families are your avatar guest it will be great. 
Like @Bronwyn100 @suggested having twins that can be put together will also offer flexibility if they are all adults.

@Villa-Barbara0 The ideal, though expensive, is zip link beds. If you go this way then make sure you state what the standard config will be (I suggest 2 single beds) and then what a guest can request with a few days notice.

@Villa-Barbara0< What type of windows and security are in the Villa?

Food for thought especially if hosting children and the legal requirement to be actively supervising them including on holidays, think of what may potentially occur if a child wandered off on such a beautiful island ( we visited Rhodos on our OE many years ago) and you are then implicated in what may become a stressful situation.
