Do you have smart pricing enabled? Weekend pricing doesn't work if smart pricing is enabled.
It might be worth checking that weekend pricing is still set and that it hasn't been reset by any recent website changes.
If it is still set and smart pricing is not on then you could try unlisting your listing for 10 minutes and then listing it again.
Some have reported that this sometimes resolves configuration issues.
If that doesn't work then contact Airbnb.
