2nd listing

Hi! I made a 2nd listing for a basic room that currently I only want to rent out to one person. (She's a travel nurse.) I'm going through and  locking every day she does not need since I don't want his listing to pop up on any other's search. Is there a way I can simply "turn off" the option for the room and turn it on when she needs to book?

You can set the listing in a way it blocks all days by default. Then you manually have to set those wanted days to "available". Which is more secure then the other way round, as the "booking window" will also not open blocked days in future.

\every  listing has a direct link with a unique id-nr, for example like one of yours: https://www.airbnb.com/rooms/23696389, she can bookmark the link for direct access in future. You can also snooze a listing, then it will not show up in search results for somebody. I don't know if she can book if listing is snoozed, you will have to give it try to find out.
