You can definitely set separate weekday and weekend limits however, what you want to do cannot be done in a completely automated manner. Here are some thoughts:

You will set the regular settings to the weekly constraints you want and then reset the weekend ones to single night stay options. In doing this, however, the "Prep Time" you set for during the week will apply, unfortunately, for during the weekend as well, so you won't get the back to back reservations you want.
Alternatively, you can manually block off the night before and after each reservation once they are made so you have the one day turn around you want. This will implement the 2-night weekday minimum and then you simply do not block off the weekends you have set to a 1 night minimum.
Mathmatically, because my mind goes to those places, what you are looking for is only 1 reservation a week (one day prep before they arrive and one day prep after they leave and 2 night minimum). This means that from Sunday to Thursday - because you book NIGHTS not days - you can take no more than 1 reservation if Friday is the beginning of the weekend. (calculating this to make sure I have that right). That being said, you could just manually block the days you don't want to turn the room based on your personal schedule and take guests that only fit your hosting availability.
