If you already have a confirmed booking for a weekend in October, and then receive a new request for a longer stay from September 21 through October 31, how would you handle the request?

That new booking request can never have been received by you , as not all dates are available.
So the question is fictional

just confirm with them which dates are available but most likely they won't want to book if you can't offer the whole time
the professional and decent thing to do is not to contact the original guest at all - I would not ever ask them to change their arrangements.

For you to have received such a request from a guest, they would have had to send you an information request as it's impossible for a host to receive a trip request for dates already booked by someone else. These are two different things with different actions to take by the host.
Look up on Airbnb the fines and other serious consequences when hosts cancel an existing reservation for no valid reason.
I would NEVER cancel an existing reservation - the cancelled dates get blocked by Airbnb anyway so you wouldn't be able to accept a longer reservation anyway!
