My listing states no smoking and a fine will be accessed. What do I do about smoking? How is a fine collected?

You can claim the guest through to the Resolution Center.
If you’re unable to reach an agreement, you can ask Airbnb to make the final decision 72 hours after the request was opened.

@Keba3 Stating in your listing that "a fine will be assessed" might help you deter smoking, but in reality Airbnb does not enforce penalty fines on hosts' behalf. Also, Airbnb does not actually collect the so-called a "security deposit," so in practice nothing is forfeited.
Instead, if you want to charge for damage or excess cleaning after the stay, you have to send the guest a Resolution request, and if they refuse the charge, Airbnb has final say over the amount you will be compensated. In most cases they'll require receipts and evidence of physical damage. Claims for smoking violations aren't always successful, and when they are, the settlement tends to be something around $50.
If you find that a guest is breaking your rules during the stay, your primary recourse is to kick them out; in most cases, they'll be refunded for the unused nights. If you want to be able to actually charge the fines and "inconvenience fees" listed in your House Rules, unfortunately Airbnb is not the platform for you.
Another thing related to your rules: you say
You must be 25yrs of age to book and stay. if under 25, the stay has to be approved by host and the host will remain on property during your stay

This is just horrible. I am having the same situation of guests smoking indoors and leaving trash all over. As well as they brought a pet I didn't know about who peed on my sofa.
The airbnb claim resolution is a nightmare. I do want to penalize for smoking @$500. It's too much stress.
A host told me you can get bigger reimbursement from airbnb if you have an invoice for the cleaning from the maid.
