booking two guests on same day

I would like to have guests check out at 12PM and then have new guests check in at 4PM the same day. How do I do that?

Set your checkout time at 12pm and your checkin time from 16pm. If they already booked, let them know when you can receive them

This is standard practise and how most hosts operate (unless they have specified in their account set  up to leave a number of days between bookings). Make sure your guests leaving in the morning are aware what time they must  check out...so that you have enough time to clean, etc
