I have a guest wanting to book and he will book tomorrow for 2 months. His request expired and another guest sent a request to book for 1 week during the time the first guest wants. I have already told the 2nd guest that someone else asked before they did. If I decline the 2nd guest, will that go against my super host status? Are the dates blocked still from the 2nd guest?

You can also ask the 2nd guest to withdraw the request. If you decline, the dates will be free for another booking. If you decline too much requests, it will effect your acceptance rate in a negative way, risking listing to be paused..But this rate is not measured for Superhost status. If you let requests expire often, it will effect your responsrate in a negative way (and brings Superhost status under pressure....)
