My house is listed as a 2 bedroom with the second smaller bedroom using a full size air mattress. I ahve had to replace the air mattresses a few times as they randomly get holes (even the more expensive ones). Are there any comfortable options that will fit in a small bedroom but can be stored in a closet so that I am not constantly buying new mattesses?
You could get a bike patch kit - my husband has fixed ours with a bike patch kit. I guess still a hassle to repair it, but then you wouldn't have to replace it?
That's true. It's just difficult to find the holes as they are so small sometimes. I will look into it.
Here's a video about finding the holes and using the patch kit. He talks a lot but you can fast forward. We just repaired this ridiculous REI mattress that retails for like $250 and it is good as new - we've used it about 7 times since patching it no problems. I looked at your picture and that is indeed a small room, so air mattress is probably a pretty decent way to go unless you want to move your office elsewhere.

A fold up Murphy bed. Also much more comfortable
There are sideways (lengthwise, that is) Murphy beds that work fairly well. They're expensive, but I think there are companies manufacturing them with plywood, which is paintable, and so you can easily get rid of the bed when not in use.
